Synthesis and antikinetoplastid activity of a series of N,N′-substituted diamines
Graphical abstract
A series of N,N′-substituted diamines were prepared. The most potent compounds have 50% parasite growth inhibition at submicromolar (against Trypanosoma cruzi) or nanomolar (against Trypanosoma brucei and Leishmania donovani) range.

A minimalistic approach to develop new anti-apicomplexa polyamines analogs
2018, European Journal of Medicinal ChemistryCitation Excerpt :The library has two sources of diversity: 1) the carbon size chain, which brings flexibility and more grades of freedom and 2) the substituent on the aromatic ring. The library was prepared in a single step by a double reductive amination between the diamines and the benzaldehydes using the procedure by Phanstiel and co-workers [23] with minimal modifications (Scheme 1). The library was built with all the possible combinations of commercial aliphatic diamines with a 3, 4, 6, 8, 10 and 12 carbon chain length and five aromatic aldehydes (benzaldehyde, 4-methoxy-benzaldehyde, 4-benzyloxy benzaldehyde, 3-hydroxy-4-benzyloxy benzaldehyde and 3-methoxy-4-benzyloxy benzaldehyde).

The fact that the most charged species 7 (see Fig. 1), probably by lipophilic compound, is also the least toxic in accordance with the findings of Caminos et al.32 who reported that the growth inhibitory activity in T. cruzi, T. brucei, and L. donovani by a series of diamine derivatives improved with increasing lipophilicity. Differences in the clinical manifestations and the low cure rate are due to high genetic variability of T. cruzi.3

Recently, Caminos et al. [90] prepared twenty-five symmetric N,N′-benzyl substituted diamines (Fig. 18). The length of the linear chain varied from 3 to 10 carbons and the benzyl moieties were mostly substituted by methoxy and benzyloxy groups.
Novel therapeutic strategies for treatment of visceral leishmaniasis
2013, Drug Discovery TodayCitation Excerpt :Nitroquiolines are one of the members of this class which showed leishmanicidal activity, and 2-hydroxy-8-nitroquinoline showed the best antileishmanial activity among some 2-substituted nitroquinolines synthesized and examined by Paloque et al. [14]. Caminos et al. [15] synthesized a library of 25 N,N′-substituted diamines and found that a diamine containing 4-O-Benzyl substitution showed the best in vitro antiparasitic activity against the microbes causing Chagas’ disease (Trypanosoma cruzi), visceral leishmaniasis (L. donovani) and human African trypanosomiasis (Trypanosoma brucei) [15]. The diamine derivatives of bisnaphthalimidopropyl (BNIP), a hydroxynaphthoquinone antiprotozoal drug, buparvaquone and compounds belonging to the class of arylimidamide have been also found to exert antileishmanial activity [16–19].
